Abstract:
Dendritic cells (DCs) are key to initiate anti-tumour immune responses, but their function is commonly disrupted in cancer. We performed a genome-wide arrayed CRISPR-Cas9 screen in a cancer cell – DC co-culture system and uncovered genes which upon knockout in cancer cells could lead to activation in DCs. Using transcriptomics together with binding partner analysis we discovered that overexpression of a single protein (LIGHT) in cancer cells could activate dendritic cells and promote T-cell activation in a cross-priming assay.
